Documentation accompanying research peptides usually includes a certificate of analysis listing the batch number, purity figure, and test methods applied. Buyers comparing suppliers should check whether the reported purity refers to chromatographic area or to a mass-balance calculation, because the two are not equivalent. Counter-ion content, residual solvents, and water content are sometimes omitted from such certificates even though they affect the actual peptide mass present. Independent verification through a second laboratory is the most direct way to confirm that a supplied material matches its label.
Purity assessment of peptide reagents normally relies on reversed-phase high-performance liquid chromatography. A gradient of acetonitrile in water with trifluoroacetic acid, paired with a C18 stationary phase, separates the target tetrapeptide from truncated sequences and deletion analogues. Detection at 214 nanometres exploits absorbance of the peptide backbone, since the molecule contains no aromatic residue. Results are expressed as a percentage of total peak area. Values above ninety-five percent are typical for research-grade material, although reporting conventions vary between suppliers.
Because epitalon has no pharmacopoeial monograph, quality assessment depends on supplier documentation and independent testing. Certificates of analysis typically report a purity figure from a single chromatographic run, a measured mass and sometimes an appearance description, but methods and acceptance criteria are not harmonised across vendors. Third-party laboratories can repeat identity and purity measurements, and mismatches between labelled and measured peptide content have been described for research peptides generally. What constitutes adequate identity confirmation for a molecule of this size stays an open question, since mass agreement alone does not separate closely related sequences.
Identity testing for epitalon relies on reversed-phase high-performance liquid chromatography for purity and mass spectrometry for mass confirmation. Because the sequence contains no tryptophan or tyrosine, ultraviolet detection at 280 nanometres is insensitive, so chromatographic methods usually monitor absorbance near 214 nanometres, where the peptide backbone absorbs. Electrospray ionisation or matrix-assisted laser desorption/ionisation then checks the intact mass against the expected value near 390 daltons. Peptide mapping or amino acid analysis after acid hydrolysis can supplement these measurements, although such confirmatory work is seldom reported on commercial certificates of analysis.

Epitalon therapy in patients with degenerative retinal lesions results in a positive clinical effect in 90% of the cases. The analysis of Epitalon effects suggests that the tetrapeptide participates in the mechanisms of transcription common for the epiphysis and retina.

The study shows that keeping rats in different light modes has a different impact on pancreatic and intestine amylase activity and has its peculiarities in regard of age as well as geroprotectors' influence on it. Melatonin use entails pancreatic amylase activity increase in mature animals in natural light of the North-West of Russia (NL) and 24-hour constant light (LD) and on the contrary amylase activity drops in case of using epitalon. Simultaneously, the enzyme activity in young and old rats under LD, melatonin and epitalon decreases. Intestine amylase activity influenced by both preparations rises in NL and drops in LD mode with respect to the control group.

[Comparative study of the effects of melatonin and epitalon on the protracted memory under the shuttle labyrinth test conditions in rats in the course of aging]. [Article in Russian] Vinogradova IA. The influence of the chronic administration of melatonin (epiphyseal hormone) and epitalon (a synthetic tetrapeptide increasing melatonin production) on the learning process and the protracted memory has been studied in LIO rats in the course of aging for 2 years under standard illumination regime (12L :12D). The daily administration of melatonin (Sigma, USA) with drinking water (in 10 mg/liter dose at night) in rats beginning with the age of 4 months did not influence the learning processes in young and adult animals but it was found to contribute to optimization of the brain cognitive function in rats in the course of aging, by improving the protracted memory process. Epitalon administered in a daily dose of 0.1 microg per animal beginning with the age of 4 months showed mnemotropic properties (decreasing the extent of memory disorders) in old rats under conditions of the shuttle labyrinth test.

Neonatal hypophysectomy in chicken, as well as that in old hen has been established to entail in 1,5 months after surgery cellular and humoral immunity disturbances, pronounced hypercoagulation and fibrinolysis depression. Administration of Epitalon (Ala-Glu-Asp-Gly) to a large extent eliminates revealed shifts. This effect appeared to be stronger in neonatally hypophysectomized chicken than in old hens.

In investigations carried out with organotypic culture of mediobasal preoptic area (MPA) of hypothalamus it was established that pineal peptide preparations epitalon (2 ng/ml) and epithalamin (100 ng/ml) stimulate the development of proliferative activity of explants in 3 month and 24 months female rats. It has been shown that epithalamin is more effective in young rats in comparison with old animals, but epitalon as well as epithalamin have almost the same less pronounced inducing effect on growth zone in MPA explants from young and old animals. This effect is tissue specific and could be dependent on inhibition of apoptosis.

RESULTS: Epitalon was proven to significantly stimulate melatonin synthesis in senescent monkeys in the evening, thereby normalising the circadian rhythm of cortisol secretion.

The effect of synthetic peptide bioregulators (Epitalon, Livagen and Vilon) on structural and facultative heterochromatin of cultivated lymphocytes have been studied among old (75-88yr.) people. The data obtained indicate that epitalon, livagen and vilon: 1) activate synthetic processes, caused by reactivation of ribosomal genes as a result of deheterochromatinization (decondensation) of nucleolus organizer regions; 2) induce unrolling (deheterochromatinization) of total heterochromatin; 3) release genes repressed by heterochromatinization (condensation) of euchromatic regions forming facultative heterochromatin; 4) epitalon and livagen induce deheterochromatinization (decondensation) of pericentromeric structural heterochromatin of the chromosomes1 and 9. However, vilon does not induce deheterochromatinization of pericentromeric structural heterochromatin. These results indicate that peptide bioregulators Epitalon, Livagen and Vilon cause activation (deheterochromatinization) of chromatin in lymphocytes of old individuals.

In the course of aging both monkeys and people reveal decreased night and average daily level of melatonin in the blood plasma and reduced hormone circadian rhythm amplitude, which evidence the disorder of the pineal gland melatonin releasing function. Peptide preparations of the pineal gland (Epithalamin--a complex of peptides isolated from the pineal gland and Epitalon--synthetic tetrapeptide) recover night release of endogenous melatonin and lead to the normalization of the hormone circadian rhythm in the blood plasma. In elderly people Epithalamin and Epitalon modulate pineal gland functional state: people with pineal gland functional insufficiency report an increase of night melatonin level. The preparations of the pineal gland, effectively increasing melatonin concentration and having no side effects, can be used in clinical geriatric practice.

Experimental data on the effect of peptides--Vilon (Lys-Glu) and Epitalon (Ala-Glu-Asp-Gly)--on induction of urinary bladder tumors in rats are presented. Treatment with Vilon was followed by a significant fall in tumor incidence in 56% of experimental animals, as compared with 75.5% in control, as well as inhibition of early-onset neoplastic changes in the bladder mucosa. No inhibitory effect of Epitalon was recorded.

Biological effects of short regulatory peptides, pinealon, vesugen, vilon and epitalon were studied in model experiments in vitro. These peptides were found not to demonstrate direct antioxidant activity but be able to restrict lipid peroxidation of human lipoproteins by modification of their structure. The short peptides increase stability of red blood cell membranes toward osmotic hemolysis. They also elevate the stationary level of intracellular reactive oxygen species and at the same time decrease (all excepting epitalon) percent of dead cells in neuronal population. The suggestion was made that under in vivo conditions, short peptides may participate in apoptosis/necrosis regulation.

Reversed-phase high-performance liquid chromatography with ultraviolet detection at 214 nanometres is the usual approach. Peak area percentage provides a purity figure relative to other components in the sample. Mass spectrometry is then applied to confirm identity.
Aspartate can cyclise into a succinimide intermediate that later hydrolyses back as isoaspartate. Both forms share the same molecular mass, so mass spectrometry alone cannot tell them apart. This shapes how purity data should be interpreted.
Freeze-dried material is normally kept desiccated below minus twenty degrees Celsius and shielded from light. Solutions are prepared shortly before use and held cold. Repeated thawing is avoided because it accelerates aggregation and degradation.
The molecule lacks aromatic residues, so it absorbs weakly near 280 nanometres, the wavelength applied to many other peptides. The peptide bond absorbs strongly below 220 nanometres, making 214 nanometres a practical compromise. Gradient methods must therefore use mobile phases with low ultraviolet absorbance to keep the baseline clean.
